چکیده

THE Gunn rat is an animal model for Crigler-Najar syndrome. a rare form of congenital unconjugated nonhemolytic hyperbilirubinemia due to the absence of glucoronyl transferase. I.e It leads to deposition of bilirubin in the basal ganglia. culminating in precocious death of the patient. Because the uptake. conjugation. and excretion of bilirubin by hepatocytes involves specific carriers and enzymes. this animal model provides a unique opportunity to investigate the interactions between proteins of donor origin with those of the recipient:' In this study we have assessed whether liver xenotransplantation can reverse the congenital hyperbilirubinemia in the Gunn rat.

Correction of congenital hyperbilirubinemia in homozygous Gunn rats by xenotransplantation of hamster livers.

The homozygous Gunn(j/j) rat is an animal model for Crigler-Najjarsyndrome in which the lack of the enzyme uridine diphosphoglucoronate-glucuronosyltransferase (UDP-GT) results in congenital unconjugated nonhemolytic hyperbilirubinemia. Because the binding of bilirubin to albumin in plasma varies from species to species, xenotransplantation (XTx) of liver afforded in this model the opportunity ...

Metabolism and disposition of C14-bilirubin in congenital non-hemolytic jaundice.

In mammals, unconjugated bilirubin is rapidly cleared from the circulation and almost quantitatively excreted as a conjugate in the bile (2-4). The efficiency and rapidity of this process indicate that conjugation and subsequent biliary excretion represent the principal pathway of bilirubin disposition. If alternate pathways of bilirubin metabolism exist under physiologic conditions, their leve...

Effect of troglitazone on the liver of a Gunn rat model of genetic enzyme polymorphism.

There is a possibility that serious liver dysfunction rarely observed in diabetic patients given troglitazone is attributable to idiosyncratic abnormalities in liver drug-metabolism. In addition, the results of blood biochemical examinations in serious cases of liver dysfunction showed a tendency for a high level of total bilirubin (T-Bil) over a long period compared with other indicators of li...

Single Liver Lobe Repopulation with Wildtype Hepatocytes Using Regional Hepatic Irradiation Cures Jaundice in Gunn Rats

BACKGROUND AND AIMS Preparative hepatic irradiation (HIR), together with mitotic stimulation of hepatocytes, permits extensive hepatic repopulation by transplanted hepatocytes in rats and mice. However, whole liver HIR is associated with radiation-induced liver disease (RILD), which limits its potential therapeutic application. In clinical experience, restricting HIR to a fraction of the liver ...

Liver repopulation with bone marrow derived cells improves the metabolic disorder in the Gunn rat.

BACKGROUND Reversible ischaemia/reperfusion (I/R) liver injury has been used to induce engraftment and hepatic parenchymal differentiation of exogenous beta2-microglubulin(-)/Thy1(+) bone marrow derived cells. AIM To test the ability of this method of hepatic parenchymal repopulation, theoretically applicable to clinical practice, to correct the metabolic disorder in a rat model of congenital...
